Hydrazide derivatives with MAO inhibitory effects (iproniazid, nialamide, isocarboxazid, etc.) are used as excellent antidepressants. Unlike INAH, these derivatives have strong toxicity and side effects, including liver cell damage, excessive excitement, orthostatic hypotension, dizziness, tinnitus, and constipation. 一般にイソニアジド INAHをさす。なお,INAHを結核患者に投与中,病識がなくなるほど気分爽快となったことから中枢神経興奮作用が研究され,モノアミン酸化酵素 (MAO) 阻害によることが明らかになった。 MAO阻害作用をもつヒドラジド誘導体 (イプロニアジド,ニアラミド,イソカルボキサジドなど) はすぐれた抗うつ剤として用いられている。この誘導体は,INAHと異なって毒性,副作用が強く,肝細胞障害,過度の興奮,起立性低血圧,めまい,耳鳴,便秘などがみられる。また,他剤との併用で過度の反応を引起すことがあるので,投与の際には注意を要する。
