Estrone - Estron (English spelling) estrone

A steroid hormone secreted from the ovaries of female animals, it is a type of follicular hormone (estrus hormone). Estrogen is a general term that includes substances with the same action, such as estradiol and estriol. It is found in large quantities in the urine of pregnant animals and was the first follicular hormone to be isolated from a natural product. It is extracted from pregnant horse urine, but estradiol, which is obtained by further reducing it, has 3 to 4 times stronger hormonal action, so estrone is used as the raw material. It is secreted from mature follicles and the placenta, and promotes the development of the gonads, sexual characteristics, and mammary glands. In particular, it works in cooperation with gonadotropic hormones secreted from the anterior pituitary gland and progesterone secreted from the corpus luteum to regulate the sexual cycle and the period from pregnancy to childbirth.

Estrone exists in equilibrium with estradiol in the blood, and is metabolized in the liver to become estriol. It is also conjugated with glucuronic acid and sulfate in the liver and excreted in the urine. A significant amount of estrone secreted in the bile is reabsorbed (enterohepatic circulation. It is absorbed from the intestine, taken up by hepatocytes and conjugated, then deconjugated by intestinal bacteria and absorbed again from the intestine).

Clinically, taking advantage of its weak biological activity, it is used as an oral tablet to improve menopausal symptoms, and as an injectable drug to induce ovulation and to stop functional bleeding (bleeding from the uterus in the absence of organic disorder).

動物の雌の卵巣から分泌されるステロイドホルモンで、卵胞ホルモン（発情ホルモン）の一種。エストラジオール、エストリオールなど同じ作用をする物質を含めエストロジェン（エストロゲン）と総称する。妊娠した動物の尿中に大量に含まれており、天然物から最初に単離された卵胞ホルモンである。妊馬尿から抽出されるが、これをさらに還元して得られるエストラジオールのほうがホルモン作用が3、4倍強いので、エストロンはその原料として使われている。成熟した卵胞や胎盤から分泌され、生殖腺(せん)の発育、性徴や乳腺の発達を促進する。とくに脳下垂体前葉から分泌される生殖腺刺激ホルモン、黄体から分泌される黄体ホルモンと共同して働き、性周期や妊娠から出産に至る期間を調節している。

　血中でエストロンはエストラジオールと平衡して存在し、肝臓で代謝されてエストリオールとなる。また、肝臓でグルクロン酸や硫酸と抱合され、尿中に排出される。胆汁中に分泌されたエストロンのうちのかなりの量は再吸収される（腸肝循環。腸管より吸収され肝細胞に摂取されて抱合を受け、その後腸内細菌で抱合体が脱抱合され、ふたたび腸管から吸収される）。

　臨床的には弱い生物活性を利用して、更年期障害の改善に経口投与錠として、また注射薬として排卵誘発や機能性出血（器質的障害がないのに子宮から出血すること）の止血用に用いられている。
