Menthol - Mentooru (English spelling)

A representative monocyclic monoterpene alcohol. Generally speaking, menthol refers to l-menthol obtained from peppermint oil, but chemically it is a collective term for 12 isomers. Natural and synthetic l-menthol and d-menthol have the cool flavor peculiar to mint, while other isomers do not. Natural and synthetic l-menthol are chemically identical and are colorless needle-shaped crystals. Japanese peppermint oil used to supply l-menthol to meet the world's demand for peppermint, but now the market is dominated by peppermint from Brazil and China and synthetic products produced in Japan, Germany, and the United States.

There are three typical methods for producing l-menthol:

(1) Manufacturing method from d-citronellal When d-citronellal obtained by fractional distillation of citronella oil is added with Lewis acid and heated, it cyclizes to become isopulegol. When a solvent is added to this and it is separated by cryogenic separation, pure l-isopulegol crystals can be obtained in good yield. When this is hydrogenated under high pressure using a nickel catalyst, pure l-menthol can be obtained in good yield.

(2) Production method from thymol: Thymol is catalytically hydrogenated to produce d- and l-menthol, which is then isomer-separated and optically resolved to produce pure l-menthol.

(3) Asymmetric synthesis method When myrcene is reacted with lithium and diethylamine, diethylgeranylamine is produced. When this is subjected to asymmetric isomerization using a (S)-BINAP-Ru complex catalyst, citronellal enamine is produced, which is hydrolyzed to pure d-citronellal. This is ring-closed with zinc bromide to produce l-isopulegol, which is then hydrogenated to produce l-menthol. l-Menthol is used in medicines (patches, ointments, aerosols), tobacco, toothpaste, hair growth products, chewing gum, candy, ice cream, beverages, cosmetics, etc.

単環性モノテルペンアルコールの代表的なもの。一般に、メントールといえば、はっか油から得られるl-メントール（はっか脳）をさすが、化学的には12個ある異性体の総称である。ハッカ特有の冷涼な香味を有しているのは天然および合成のl-メントールとd-メントールで他の異性体は冷涼な香味を有していない。天然はっか脳と合成のl-メントールは化学的には同一で、無色の針状結晶である。日本産和種はっか油はl-メントールの供給源として世界のハッカ需要をまかなっていたが、現在はブラジルおよび中国産はっか脳と日本、ドイツ、アメリカで生産される合成品が市場を支配している。

　l-メントールの代表的製法は次の3種類である。

(1)d-シトロネラールからの製法　シトロネラ油の分留で得られるd-シトロネラールにルイス酸を加えて加熱すると環化してイソプレゴールとなる。これに溶剤を加えて深冷分離すると純粋なl-イソプレゴールの結晶が収率よく得られる。これをニッケル触媒を用いて高圧水添すると、純粋なl-メントールが収率よく得られる。

(2)チモールからの製法　チモールの接触水添によりd、l-メントールとし、異性体分離、光学分割を行い、純粋なl-メントールとする。

(3)不斉合成法　ミルセンにリチウムとジエチルアミンを反応させると、ジエチルゲラニルアミンができる。これを(S)-BINAP-Ru錯体触媒を用いて不斉異性化反応を行うとシトロネラールエナミンを生じ、これを加水分解して純粋のd-シトロネラールとする。これを臭化亜鉛で閉環し、l-イソプレゴールに導き、水素添加によってl-メントールを製造する。l-メントールは医薬品（貼(は)り薬、軟膏(なんこう)、エアゾール）、タバコ、歯磨剤、育毛剤、チューインガム、キャンディー、アイスクリーム、飲料、化粧品などに用いられる。
