Semen diluted with a preservation solution and frozen in liquid nitrogen (-196°C). Used for artificial insemination of livestock. When livestock sperm is diluted with a preservation solution mainly made of egg yolk, sodium citrate, and glucose and stored at around 5°C, it can only retain its fertilizing ability for 4 to 5 days. However, in 1952, C. Polge and his colleagues in the UK succeeded in freezing semen on dry ice (-79°C) using a preservation solution containing glycerin as a cryoprotectant (7% of the total volume of diluted semen), and this was followed by a shift to cryopreservation.
