A special cytochemical staining method used to clarify the type and function of white blood cells. Peroxidase is an enzyme that activates the oxygen atom of hydrogen peroxide to oxidize a substrate. The peroxidase reaction utilizes the principle of this enzyme reaction to stain the site where peroxidase is present using a dye that changes color when oxidized to a substrate. There are various types of normal white blood cells, but among these, granulocytes and monocytes, which have phagocytic and bactericidal functions, contain peroxidase in their cytoplasmic granules (positive peroxidase reaction), and their oxidative action enhances their bactericidal ability. 白血球の種類や機能を明らかにするために用いられる細胞化学的特殊染色方法の一つ。ペルオキシダーゼは過酸化水素の酸素原子を賦活して基質を酸化する酵素であるが,この酵素反応の原理を利用して,基質に酸化されると発色する色素を用い,ペルオキシダーゼの存在部位を染め出すのがペルオキシダーゼ反応である。正常な白血球にはいろいろな種類があるが,このうち貪食・殺菌能をもつ顆粒球,単球は,細胞質顆粒中にペルオキシダーゼを有し(ペルオキシダーゼ反応陽性),その酸化作用によって殺菌能を増強する。
