If an aqueous solution of sodium hydroxide or potassium hydroxide is added to the sample solution to make it alkaline, and then a small amount of an aqueous solution of several percent copper sulfate is added, it will turn a bluish purple to reddish purple color. It is named after the fact that under the same conditions, biuret forms a complex and turns color. The color of proteins is also caused by a complex of similar structure. Color is observed in compounds in which two -CO-NH- atomic groups are bonded with one nitrogen or carbon atom between them.
