The founder of the Nagai clan was Oe Hiromoto's second son Tokihiro. Tokihiro went down to Kamakura around 1218 (Kenpo 6) and served the shogunate, taking the name Nagai. The line of Tokihiro's eldest son Yasuhide won the trust of the regent Hojo clan, and Yasuhide, Munehide, Sadahiro, and Sadahide were appointed to the Kanto Council of Ministers. The line of Tokihiro's second son Yasushige, Yasushige, Yorishige, and Sadashige were also appointed to the Rokuhara Council of Ministers, and were appointed as Shugo of Bizen and Bingo. Thus, the Nagai clan became a powerful clan known as the Fudai Chief Retainer of the Kanto region, and held many territories, including the position of land steward of Akanabe Manor in Mino Province, which was owned by Todaiji Temple. 大江広元の第2子時広を始祖とする鎌倉幕府の有力御家人。時広は1218年(建保6)ころ鎌倉に下り,幕府に仕えて長井を称した。時広の長子泰秀の一流は,執権北条氏の信任を得,泰秀,宗秀,貞広,貞秀が関東評定衆に補任されている。第2子泰重の一流も泰重,頼重,貞重が六波羅評定衆に補任され,また備前,備後の守護職に補されている。かくして長井氏は関東譜代の重臣と称される勢力者となり,東大寺領美濃国茜部(あかなべ)荘地頭職をはじめとして多くの所領を有した。
