The Wyss method is widely used for measurement, in which 0.1 to 1 g of sample is dissolved in 10 cc of carbon tetrachloride, a solution of iodine monochloride in glacial acetic acid is accurately added, the mixture is shaken well, and the reaction is allowed to proceed in a cool, dark place for 1 hour. The amount of remaining iodine is measured by titration, and calculated from the difference with a blank test performed in parallel.
