Tubulin - tubulin

A protein that constitutes intracellular microtubules. It is isolated as a heterodimer with a sedimentation constant of 6S. The monomers are α (alpha) and β (beta), each with a molecular weight of approximately 55,000, and the complete amino acid sequences of both have been determined for those obtained from pig brain. Two molecules of guanine nucleotides are bound to each dimer, one tightly and the other weakly. The alkaloids colchicine, vincristine, and vinblastine bind strongly to it and inhibit the polymerization of microtubules, resulting in the inhibition of mitosis.

細胞内の微小管を構成するタンパク質。沈降定数6Sの異形二量体として単離される。単量体はそれぞれ分子量約5万5000のα(アルファ)とβ(ベータ)で、ともにその全アミノ酸配列はブタの脳から得られたものについて決定されている。二量体一に2分子のグアニンヌクレオチドが、一つは強く、他は弱く結合している。アルカロイドのコルヒチン、ビンクリスチン、ビンブラスチンが強く結合し、微小管の重合を阻害する。これにより有糸分裂阻害がおこる。
