Reverse transcriptase

An enzyme that synthesizes complementary DNA using RNA as a template. It is also called RNA dependent DNA polymerase. While the synthesis of RNA using DNA as a template is called transcription, this enzyme synthesizes DNA using RNA as a template, so it is called reverse transcription.

Some RNA viruses that cause tumors retain their genetic information in the host for a long time after infection, and H.M. Temin claimed that the RNA is copied into DNA. In 1970, H.M. Temin and D. Baltimore independently discovered that the virus particles themselves have reverse transcriptase activity, and were awarded the Nobel Prize in Physiology or Medicine in 1975 for this discovery.

After viral infection, this enzyme synthesizes complementary DNA (cDNA) to the RNA in the virus particle, first creating an RNA-DNA hybrid, then the RNA is degraded by the RNase H activity (which degrades only RNA that is base-paired with DNA) contained in the enzyme, and finally the double-stranded DNA is formed by the DNA polymerase activity, which is integrated into the host chromosome. It was initially thought to be found only in cells infected with RNA viruses, but has since been found in normal cells, and it is thought that it may reverse transcribe normal messenger RNA (mRNA) into DNA. It is one of the enzymes often used in gene manipulation experiments to synthesize DNA (cDNA) corresponding to mRNA. It is considered important because it demonstrated the reverse flow of genetic information (RNA → DNA) compared to the central dogma proposed by F.H.C. Crick, which is DNA → RNA → protein.

RNAを鋳型templateとしてそれに相補的なDNAを合成する酵素。RNA依存性DNAポリメラーゼ（RNA dependent DNA polymerase）ともよばれる。DNAを鋳型としてRNAを合成するのを転写transcriptionというのに対して、この酵素はRNAを鋳型としてDNAを合成するので逆転写という。

　腫瘍(しゅよう)をおこすRNAウイルスには感染後長い期間宿主中にその遺伝情報を残すものがあり、H・M・テミンはRNAがDNAにコピーされていると主張した。1970年にH・M・テミンとD・ボルティモアはおのおの独立にウイルス粒子自体に逆転写酵素活性があることを発見し、この業績により1975年ノーベル医学生理学賞を授与された。

　ウイルス感染後、この酵素はウイルス粒子内にあったRNAと相補的なDNA（complementary DNAといい、cDNAと略記）を合成、いったんRNA‐DNAハイブリッド（hybrid雑種）がつくられ、次にこの酵素中に含まれるリボヌクレアーゼRNase H活性（DNAと塩基対をつくっているRNAのみを分解する）によりRNAが分解され、最後にDNAポリメラーゼ活性により2本鎖のDNAとなり、宿主染色体中に組み込まれる。最初はRNAウイルス感染細胞にのみあると思われていたが、その後正常細胞からもみつけられており、通常のメッセンジャーRNA（mRNA）がDNAに逆転写される可能性も考えられている。遺伝子操作実験ではmRNAに対応するDNA（cDNA）を合成するためによく使われる酵素の一つである。F・H・C・クリックが提唱したDNA→RNA→タンパク質という遺伝情報の流れ、いわゆるセントラルドグマに対して、RNA→DNAという逆向きの流れを示したとして重要視されている。
