Radioimmunoassay

This method was developed in 1960 by RS Yalow and SABerson as an immunoassay for plasma insulin. It is also called radioimmunoassay. The principle is that when a certain amount (T) of an antigen (e.g. insulin) labeled with a radioisotope is reacted with a certain amount of an antibody produced by immunizing a different species of animal with the antigen, a labeled antigen-antibody complex (B) is produced, and the remaining labeled antigen remains free (F). When unlabeled antigen (antigen standard or sample containing antigen) is added to this system, it and the labeled antigen compete with each other to bind to the antibody, and an equilibrium state is reached.

1960年，ヤローR.S.YalowとバーソンS.A.Bersonにより血漿インシュリンの免疫測定法として開発された方法。放射免疫測定法ともいう。原理は，放射性同位元素(ラジオアイソトープ)で標識した抗原(たとえばインシュリン)の一定量(T)に，その抗原で異種動物を免疫して作った抗体の一定量を反応させると標識抗原‐抗体結合物(B)を生じ，残りの標識抗原は遊離の状態(F)で残る。この系に非標識抗原(抗原の標準品または抗原を含む試料)を加えると，これと標識抗原は競合しながら抗体と結合し，平衡状態となる。
