Urobilin

C 33 H 42 N 4 O 6 (590.72). Also called mesobin. A brown bile pigment present in feces and urine that is Ehrlich reaction negative, it is known as urobilin bodies together with urobilinogen. It is produced when the bile pigment bilirubin is reduced to methobilirubin, which is then oxidized to produce colorless urobilinogen. In feces, it exists mostly in the form of urobilinogen. Although only trace amounts are present in normal urine, excretion increases in some cases of liver dysfunction, and therefore quantification of urobilin bodies is used as a sensitive liver function test. Urine turns brown when left to stand because urobilinogen is oxidized by air to form urobilin.

Obtained as needle-shaped crystals from chloroform. Melting point 177 °C. Soluble in ethanol and chloroform, sparingly soluble in ether and water. λ max 330, 452 nm in dioxane. Reacts with zinc acetate in ethanol, emitting strong green fluorescence (λ max 509 nm, Schlesinger reaction), distinguishing it from urobilinogen. It also reacts with copper(II) ions and mercury(II) ions, producing a red color. [CAS 1856-98-0]

C33H42N4O6(590.72).メゾビン(mesobilin)ともいう.糞尿中に存在し,エールリヒ反応陰性な褐色胆汁色素で,ウロビリノーゲンとともにウロビリン体とよばれる.胆汁色素ビリルビンが還元され,メソビリルビンを経て無色のウロビリノーゲンが生成し,これが酸化されて生じる.糞中ではほとんどがウロビリノーゲンの形で存在する.正常尿には微量しか存在しないが,ある種の肝臓機能障害などの場合には排出量が増加するので,鋭敏な肝機能検査法としてウロビリン体の定量が用いられる.尿を放置すると褐色になるのは,ウロビリノーゲンが空気酸化されてウロビリンになるためである.

クロロホルムから針状結晶として得られる.融点177 ℃.エタノール,クロロホルムに可溶,エーテル,水に難溶.ジオキサン中のλmax 330,452 nm.酢酸亜鉛とエタノール中で反応し,強い緑色蛍光を発する(λmax 509 nm,Schlesinger反応)ことでウロビリノーゲンと区別される.ほかに銅(Ⅱ)イオン,水銀(Ⅱ)イオンとも反応し,赤色を呈する.[CAS 1856-98-0]
